Operation VBCF PlantS facility

Dear all,
due to the given design and setup of the Facility (remote control) and taken measures we will be able to keep up the main operation of the phytotrons and most of our (digital) services (…from today’s point of view).
Please find important information, taken measures, reaction times and contact details summarized below:
1. We have increased the stock of consumables to avoid ordering and deliveries in the upcoming 3-4 weeks.
2. Anneliese and Jolanta will reduce their on-campus presence to a necessary minimum adapting to the actual workload of the research groups
3. Phytotron operation and control incl. alarms and repairs will continue via remote control/home-office mode by Anneliese and Jakub
4. Maintenance and repairs will continue, as usual 24/7, by our external partner Incotec-Cooling GmbH and GMI/IMBA Haustechnik
5. Image- and data analysis service (Ethan) as well Software development & PHENOBox (Sebastian) will continue in home-office mode
6. Please direct your requests, as usual, to: plants(a)vbcf.ac.at<mailto:plants@vbcf.ac.at>; in urgent cases (additionally to the mail!) call 1. Jakub: +43664 8084 77 090 and/or 2. Anneliese: +43664 80 51 53 9101

Waste Bins in PlantS Facility

Dear all,
a short reminder of "General Plant Rules":
Do not fill the waste bins with orange autoclave bag more than half full. Full bins with wet soil are too heavy for our waste management staff!
And do not collect waste in orange autoclave bags outside of waste bins, please ask me for extra bins if you need them.

CO2 atmosphere control @PlantS & GMI GeneGun move to MFPL

Dear all,
We have completed the installation of the CO2 control system in phytotron #22 and #23 (MFPL location).
CO2 concentrations of ambient/400ppm up to 2000ppm can be simulated with a precision of +/- 5ppm.
Higher values can also be set upon special request.

The GeneGun, which is currently located @GMI room #8.18, will be moved tomorrow morning to the MFPL PlantS Lab.

HT Phenotyping Phytotron available!

Our high-throughput plant phenotyping phytotron is again available from February on till end of April!

This particular phytotron is capable of providing various abiotic stresses like cold (-15C), drought, waterlogging, variable spectral light conditions (LED) and complex environmental simulations (dynamic simulations of temperature and rel. humidity).
We provide full HT phenotyping service – from the seed via image analysis to data analysis and statistics.

Dear Peter, Dear all,
Thank you for your input; I appreciate the open discussion.
That basically confirms our findings and the literature: There is a soil/nutrient AND a light component that is causing the stress; but the one is probably linked to the other.
Please refer to the publications and references listed in our last slide.
Our hypothesis also considers both factors; but what it further proposes is, that if we fix the soil/nutrient issue the light factor has no, or at least a not significant, effect (still under deeper investigation but proofed by our first experiment).
Do you agree?
And again, the most stressed plants in our experiment were grown under fluorescent tubes.
There was not one stressed plant at any light conditions in all the fertilized samples.
Your experiment proofs the high variability of results under suboptimal soil/nutrient conditions (remember the “jumping bad-guy and good guy” game).
And YES; the LED light is designed to be closer to the natural sunlight and is therefore most probably more “powerful” than the artificial fluorescent tube spectrum; the more we should take care on the on the factors.
Important remark to the experiment setup in respect to this and future experiments:
Based on the images in your presentation (slide 11 to 14) it is very difficult, or even impossible to evaluate or judge, because of the different light conditions that were used for imaging!
One either needs to take all the samples out of the phytotron to uniform light conditions for imaging (like e.g.we did, ring-light is available at our Facility), or use a color standard (available in our Facility) and correct for the light in a post-process manner. Otherwise the green color of the plant will always appear different (good/bad) in different setting although it is same colored; even the tray per se looks stressed under LED light (bluish/reddish).
Therefore, we are happy to collaborate and be more involved in future experiments.
More tests (by PlantS) are planned for 2020 and we will keep you up to date; we will also arrange a user meeting February/March.

in addition to the VBCF tests sent by Jakub the other day (many thanks!) we also have some interesting additions to make. Please refer to the attached pptx.
As you will see, when using the soil from Gramoflor without any additional fertiliser but with fluorescent tubes, plants are perfectly fine during their life-time.
The same soil, without fertiliser, leads to severe stress symptoms under LED lights.
It is important to note, that plants grown on this non-fertilised soil and fluorescent tubes but exposed to stray light from LEDs develop stress symptoms (not so severe as under direct LED light, but still). This latter point also helps to straighten some ambiguity with respect to some earlier tests made with fluorescent tubes.
In this sense, we need to consider two stress factors: light emitted by the LED lamps and sub-optimal soil.
The pptx also contains some pictures testing the former ED63 soil and taller pots.
We will now evaluate if the "double saved plants” (fertilised soil and fluorescent tubes) perform better for us (meiosis!) than the “single saved plants” (fertilised soil and LEDs).

Irrespective of the spectral conditions, most of the untreated plants show, to a certain extent, stress symptoms and Anthocyanin accumulation.
This applies also to the fluorescent tube control spectrum (!). In this particular experiment the fluorescent tube control shows even the strongest stress symptoms.
On the other hand, ALL treated (fertilized) samples look very healthy and do not show any stress symptoms.
This quick test and the literature listed seems to proof our hypothesis: “Photooxidative stress caused by nutrient (Pi?) deficiency”.
We will continue with the investigation focusing on the soil and nutrients.

In the last weeks/months we have been investigating the plant stress issue, mainly focusing on the LED light spectrum.
Always comparing to the fluorescent tube standard.
Surprisingly, also the fluorescent tube setup shows same plant stress issues!
Which was not the case in the very beginning when we have tested and decided for the new type of soil.
See picture attached to this mail:
Phytotron: #21
Soil: Gramoflor2006; not sieved; with perlite
Light: Fluorescent tubes; LD
Watering: Flooding twice a week
Left side of the tray: WUXAL treated plants (once, after 1 month, 2ml/L)
Right side of the tray: untreated plants
Our new hypothesis focuses on the soil.
In particular, on the soil composition and the available nutrients.
From today’s point of view, it looks like the composition of the soil might have changed over the last months (without informing us); but this needs further investigation.
The automated watering system (flooding) also eventually contributes negatively to the problem by washing out nutrients; twice a week.
Also sieving might be suboptimal.
Red leaves are known to be a symptom for Phosphor/Phosphate deficiency.
We are in touch with the soil supplier and with APPN soil scientist from the BOKU to find a long-term solution.
As a quick fix
* you can fertilize your plants with WUXAL fertilizer which is available in the Facility (once after ~1month; 2ml/L; watering from the top).
* Use a long-term fertilizer (OSMOCOTE)
* Use the old type of soil ED63
Please contact Anneliese in case of questions for the quick fix.
We will keep you posted on further steps and present you soon our final results and the long-term solution.

On the other hand, ALL treated (fertilized) samples look very healthy and do not show any stress symptoms.
This quick test and the literature listed seems to proof our hypothesis: “Photooxidative stress caused by nutrient (Pi?) deficiency”.
We will continue with the investigation focusing on the soil and nutrients.
A new type of soil should arrive soon for testing (details see presentation).

Dear friends,
In the last weeks/months we have been investigating the plant stress issue, mainly focusing on the LED light spectrum.
Always comparing to the fluorescent tube standard.
Surprisingly, also the fluorescent tube setup shows same plant stress issues!
Which was not the case in the very beginning when we have tested and decided for the new type of soil.

Phytotron: #21
Soil: Gramoflor2006; not sieved; with perlite
Light: Fluorescent tubes; LD
Watering: Flooding twice a week
Left side of the tray: WUXAL treated plants (once, after 1 month, 2ml/L)
Right side of the tray: untreated plants
Our new hypothesis focuses on the soil.
In particular, on the soil composition and the available nutrients.
From today’s point of view, it looks like the composition of the soil might have changed over the last months (without informing us); but this needs further investigation.
The automated watering system (flooding) also eventually contributes negatively to the problem by washing out nutrients; twice a week.
Also sieving might be suboptimal.
Red leaves are known to be a symptom for Phosphor/Phosphate deficiency.
We are in touch with the soil supplier and with APPN soil scientist from the BOKU to find a long-term solution.
As a quick fix
* you can fertilize your plants with WUXAL fertilizer which is available in the Facility (once after ~1month; 2ml/L; watering from the top).
* Use a long-term fertilizer (OSMOCOTE)
* Use the old type of soil ED63
Please contact Anneliese in case of questions for the quick fix.
We will keep you posted on further steps and present you soon our final results and the long-term solution.

Construction works @PlantS Facility GMI and MFPL

Dear all,
Next week small construction works are planned that might slightly affect the daily operation (mostly accessibility/corridor) of the Facility.
In the next weeks we will also need to shut down phytotron #22 and #23 (one by the other) for a significant amount of time.
No other phytotrons will be affected!
GMI -1: Tuesday 3rd Dec and Wednesday 4th Dec
Replacement of a pipe in the corridor (this particular pipe is not at all related to the operation of our Facility).
MFPL 8th floor: 2.5 weeks from the 2nd December on
CO2 System installation in #22 and #23

Thank you for your prompt feedback!
Well, this is certainly not all, but a (if not the) crucial factor.
We will also of course continue with LED spectra experiment and share with you the results.
There are so many factors to be considered!
We will also investigate the fluorescent tube chamber at the GMI.
Issues to consider here are: the type of fluorescent tubes (T8 GMI vs. T5 MFPL), 4 tubes or 6 tubes, watering (some users water from the top!), pot size (height, washing out of nutrients) and soil sieving.
Regarding the surrounding LED light I just want to clarify that the LEDs are giving a down pointed light with way less cross-lighting than fluorescent tubes.
The experiments are also (if possible) done I the most upper shelf unit to avoid cross-light as good as possible.
As discussed, we also could not measure any significant difference in the spectrum irrespective of the position and direction of our spectrometer.
But, yes, we will also investigate this! Thanks.

Systematic information will follow. This was just an update and quick fix mail in order to help you as soon as possible (image was taken today, same as the fluo tube image shared in the last mail!).
No stress symptoms so far; but a significant difference between treatment and control.

I am not convinced though that this is all. I would like to hear more about the GMI chamber that has exclusively fluorescent lamps installed but apparently houses plants without stress symptoms. What soil was used there?
As I mentioned to Jakub in an earlier email the plants in chamber #21 (relating to the picture) are still exposed to the light from the LEDs, though not directly.
They are grown under fluorescent tubes, but are surrounded by LEDs.
Can I/we get some more systematic information on this please.
